Puko is friends with 
Retsuko and is an island visitor. To have her as a possible visitor, the player must complete the "Everything is Fine" quest.
She will visit the player every 2 weeks, if her 
Visitor Cabin Requirements are met. Until she reaches a 
5-star friendship level on your island, she will assign Daily Quests, one per visit to your island. Her daily gift for talking to her is 1x Espresso (Item).
Check the weekly visitor schedule for details on when she visits!

Puko as a permanent 5-star resident are one of the requirements to unlock the following quests:

For Puko to move into a 
Visitor Cabin, it must meet the following requirements:
- Must be in
Mount Hothead to be near Retsuko. - Must have a
Pillow Furniture - Must have three
Table Furniture. - Must have five
Chair Furniture. - Must have a
Mirror item. - Must have a
Tropical item. - Must have a
Swampy item. - Must have a
Rocky item. - Must have a
Aquatic item.
